All limb-girdle muscular dystrophies are caused by a genetic mutation that affects the functioning of muscles. Often, the mutation causes problems with the proteins that form muscle cell membranes. These genetic problems lead to muscle weakness that most commonly affects voluntary muscles in the shoulders, hips, and thighs, but can also affect muscles in the heart. In most cases, the disease progresses over time and can lead to a loss of mobility.
