All types of limb-girdle muscular dystrophy are caused by damaged or defective genes. The particular genetic mutation that causes it depends on which type of the disease you have. Most commonly, the cause is a genetic defect that causes problems in the proteins that make up the muscle cell membranes. Because of this lack of protein, the muscle cells don't function properly. This leads to muscle weakness, especially in the shoulders and hips.
