Isentress is the first drug in a group called HIV-1 integrase strand transfer inhibitors. These drugs prevent the integrase enzyme from playing its role in inserting viral genetic material into the host genetic material. One requirement for HIV self-replication is the insertion of some of the viral DNA into the host DNA.
